Other justification: De keuze voor FANG ELISA assays is essentieel om de wetenschappelijke en regulatorische vereisten van onze studie te respecteren. De financiers (CEPI, J&J, MSD) van deze studie hebben expliciet gevraagd om FANG ELISA als primair endpoint te gebruiken. Dit is noodzakelijk om de resultaten van onze studie te kunnen vergelijken met eerdere klinische onderzoeken uitgevoerd tijdens vorige Ebola-uitbraken, waarin uitsluitend de gecertificeerde FANG ELISA werd toegepast. Deze historische gegevens vormden de basis voor FDA- en EMA-licenties van bestaande vaccins. Het gebruik van FANG ELISA in onze studie is daarom cruciaal om de mogelijkheid tot toekomstige licentiëring van de te testen boosterdosis te waarborgen. Ons goedgekeurd studieprotocol stipuleert duidelijk dat FANG ELISA wordt gebruikt voor het primaire immunogeniciteitsendpoint: FANG ELISA units/mL van anti-EBOV IgG op dag 21 na toediening van de booster. Dit is de enige methode die regulatorisch erkend is en die vergelijkbaarheid met eerdere studies garandeert. Alle andere immunologische analyses (zoals multiplex Luminex-tests) worden aanvullend uitgevoerd, maar kunnen het primaire endpoint niet vervangen. De FANG ELISA zal worden uitgevoerd in samenwerking met een gecertificeerd laboratorium, conform internationale kwaliteitsstandaarden.
