Description: 1 Stück Nanoskopie-Mikroskopsystem Für die Strukturanalyse fixierter Proben ist die optische Auflösung der entscheidende Parameter. Eine hohe optische Auflösung ermöglicht es, zelluläre Organisationen besser zu erkennen und damit den positiven wie auch den negativen Einfluss von Substanzen präziser zu beurteilen. Um detaillierte Protein- und Membranorganisationen wie Proteindimere sowie kleine Poren in Biomembranen mikroskopisch analysieren und zudem Proteinkonformationen unterscheiden zu können, ist ein Mikroskop mit einer optischen Auflösung von bis zu 5 nm in 3D oder unter 1 nm in 2D notwendig. Um die Bewegung von Proteinen oder anderen zellulären Strukturen innerhalb einer Zelle verfolgen zu können, ist es wichtig, dass sowohl der zeitliche Abstand zwischen zwei Lokalisationen so gering wie möglich als auch die Präzision der Lokalisation so hoch wie möglich ist. Insbesondere für die Analyse der Dynamik einzelner Proteine in Biomembranen ist eine zeitliche Auflösung von bis zu 1 ms sowie im Lebendzellmodus eine Lokalisationspräzision von 10 nm in 3D erforderlich. Die Fraunhofer-ITMP-Außenstelle in Göttingen hat sich seit ihrer Gründung vor fünf Jahren zu einer bekannten Adresse für moderne und hochauflösende Mikroskopietechniken entwickelt, die Messungen im Nanometerbereich nach Industriestandard ermöglichen. Infolgedessen erhalten wir vermehrt Anfragen von bestehenden und neuen Industriepartnern, ob wir Messungen mit den oben beschriebenen Parametern durchführen können; dies ist momentan jedoch leider nicht möglich. Um Anfragen aus der Industrie bearbeiten zu können und die bestehende technologische Lücke am ITMP-Standort Göttingen zu schließen, soll ein MINFLUX-Mikroskop angeschafft werden. Mit einem MINFLUX-System lassen sich dynamische Strukturen in lebenden Zellen mit einer zeitlichen Auflösung von bis zu 1 ms und einer optischen 3D-Auflösung von etwa 10 nm untersuchen. Darüber hinaus können fixierte Proben mit einer räumlichen Auflösung von unter 1 nm bis etwa 5 nm analysiert werden. Damit eröffnet die Technologie die Möglichkeit, biologische Prozesse mit bislang unerreichter räumlicher und zeitlicher Präzision zu charakterisieren und sowohl wissenschaftliche als auch industrielle Fragestellungen auf höchstem Niveau zu bearbeiten.
