To meet the requirements for isobaric tag approaches with TMT18plex, the Orbitrap technology must be used. The Orbitrap's instrument intrinsic high resolution at low mass ranges ensures accurate detection/quantification of the low mass reporter ions, which are the result of isobaric tag fragmentation in the Mass Spectrometer.
The other technology available on the market for Mass Spectrometry, TOF (Time of flight), has the opposite resolution profile, i.e. shows higher resolution at higher mass ranges but insufficient resolution at the lower mass ranges and is thus unable to resolve all the TMT18plex reporter ions. TOFs are unable to resolve the TMT18plex reporter ions due to insufficient resolution at the mass range (120-140 m/z) where reporter ions lay, and thus are not useful for isobaric tag approaches with TMT18plex.
The Orbitrap technology is unique for Thermo Scientific since it is protected by patent (US Patent 7714283).
