The Centre for Cellular Imaging would like to buy a High Pressure Freezing (HPF) system for electron microscopy to improve sample preparation/imaging workflow on Correlative Light and Electron microscopy (CLEM). HPF is a technology that aim to vitrify with liquid nitrogen hydrated specimen up to 200 μm in thickness, allowing direct cryofixation of biological samples without any chemical fixation (important for ultrastructure preservation during electron microscopy sample preparation). To achieve that, we exploit in part the water phase diagram and circumvent the ice transition by placing our object at 2 000 bars before a rapid cooling. Our facility is also interested in coupling of the HPF to our light microscopes to minimising the delay between dynamic imaging and cryoimmobilisation, in order to fix the events of interest in both space and time.
