Vanquish Horizon UHPLC- Q Exactive HF

The Department of Molecular Sciences at SLU has decided to purchase a high resolution mass spectrometer of type Q Exactive HF LC-MS for analysis of small organic molecules.

The intended small molecule analyses require an ultra-high pressure (up to 1500 bar) LC-system and a mass spectrometer with resolution up to 240 000, high mass accuracy and precision in both MS and MS/MS spectra and the ability to acquire high resolution spectra via switching between positive and negative ion detection within 1 second.

A variety of research areas at SLU (including e.g. natural products chemistry and metabolomics) rely on our expertise in using information from mass spectrometry to reveal the detailed contents of small molecules in complex biological samples. To utilize this expertise in future research we need to purchase a mass spectrometer with high sensitivity that can acquire mass spectra with high information content — i.e., mass spectra with best possible mass resolution and mass accuracy, both for intact molecular ions and their fragment ions. For example, we need to discern isotopes of the same nominal mass in tracing experiments. Analyses of complex biological samples require separation with liquid chromatography and many research projects involve thousands of samples, which makes it necessary to minimize the time for data acquisition. Both positive and negative ions must often be investigated for a comprehensive analysis of biological samples and to save analysis time and time for building libraries it is essential to have the possibility to switch polarity rapidly during analysis of each sample injected.

We therefore require a LC-MS system that can provide fast chromatography (peak durations of a few seconds) and during such a duration can perform high resolution MS (≥ 120 000 for 55 < m/z < 800) with < 1 ppm mass accuracy, high resolution MS/MS with < 1 ppm mass accuracy, a point-to-point mass precision < 2 ppm over a wide range of chromatographic peak intensities and acquisition of both positive and negative spectra at high resolution (60 000 at m/z 200) within 1 second. We found that within our budget these criteria are only fulfilled by the Vanquish Horizon UHPLC- Q Exactive HF system.

The Vanquish Horizon UHPLC- Q Exactive HF is a versatile LC-MS system for analysis of small molecules. The Vanquish Horizon ultra-high performance liquid chromatograph can due to its high pressure tolerance (1 500 bar) handle a wide range of methods including those that aim at very short analysis times. The Q Exactive HF is a mass spectrometer that combines advanced quadrupole technology with an ultra-high field orbitrap mass analyzer that allows for acquisition of high resolution mass spectra at a high rate.
