Voluntary ex ante Transparency notice
The users of the Flow Cytometry Core Facility at CMM have requirements for cell sorting at a technically very advanced level and need to detect >40 parameters (scatter and fluorochromes) with a 6-way sorting function. Because of this we require a cell sorter that is spectral and:
- Measures emitted light across the spectrum (a so-called spectral flow cytometer) with APDs or avalanche photodiodes (patent US20170307505A1) which allow for greater sensitivity especially in the red part of the spectrum thanks to the improved quantum efficiency as compared to PMTs which are used in conventional non-spectral cell sorters. Examples of low light emitters include MHCII tetramers, chemokine receptors and intracellular cytokines.
- Measures Side Scatter with a 405nm (violet) laser. This is critical when sorting small particles (nanoparticles) such as extra-cellular vesicles, as well as for discriminating debris and red blood cells.
- Has flat top optics (patent US10788411) that achieves similar CV values when sorting at low, medium or high flow rates. This is critical when sorting rare cells or cells that are fragile or “difficult”.
- Has the ability to do autofluorescence extraction for discrimination of true fluorescence emission when sorting highly autofluorescent cells.
As per the technical requirements stated in our technical motivation, the only cell sorting flow cytometer that meets them is the Cytek CS. Neither BD, Sony, Thermo Fisher, or Beckman Coulter meet these specific requirements. Specifically, BD’s 6-way full spectrum cell sorter does not have APDs or flat top optics. Sony’s cell sorter is not full spectrum, cannot sort 6-ways, nor does it have flat top optics. Thermo Fisher’s 6-way cell sorter does not have APDs or flat top optics. Beckman Coulter’s 6-way cell sorter is not full spectrum and does not have APDs.
