A fully automated laser micro dissection microscope will enable us to automatically import masks from previously detected and segmented cells of interest. The system is moving the laser, not the sample, which is extremely important for our setup. This laser will cut along the masked area of interest. It will be equipped with a variety of collecting devices, like strips of 8 wells, 96 well plate, 96 micro tube plate and the system is collecting the dissected samples via gravity, which is crucial for our workflow. Those dissected samples will then be analyzed by Mass Spectrometry. The center already has one of this microscopes, but since it´s heavily overbooked and the demand is rising steeply, we need one more to help overcome this bottleneck.

Verklaring:

Leica LMD7 moves the laser, not the sample cuts directly, real-time with ‘Move and Cut’ specimen collection by gravity. This prevents any possible contamination and stress to the sample during the removal phase collects samples into strips of 8 wells, 96 well plate, 96 micro tube plate LMD7 is the only one to use a solid-state UV laser pulse with a wavelength of 349 nm, whereby one can adjust laser power and Aperture directly during the laser cut we have setup a workflow, where an additional screening microscope and the image analysis of that data output is being used in order to define the masks where this LMD7 microscope will cut the cells of interest. This workflow is well established and being used on a daily basis in order to prepare samples for mass spectrometry. Since this is a workflow we are dependent on, we can't change the setup, so we need the very same microscope in order to be able to continue with our work routine.

Purchasing a second LMD7 microscope gives us the advantage to be able to continue with our well integrated workflow.

We are still collaborating with Leica on developing new features for this microscope, that are needed for our experiment workflow. This is crucial for moving a step further with our research and therefore it is crucial to purchase the very same microscope.

US7035004B2, JP3996773 and EP1276586B1 (effective in Germany) covers a prism device and its control unit which directs the laser beam through the objective and allows moving of the laser beam in the visual field of view while the stage is stopped.

DE10018251C2, JP4146642 and US6907798B2 relates to the LMD specific stage with receiving unit that allows to move the receiving unit and to bring only the opening of the selected receiving element into the correct catching position for catching the dissectate.

DE10057292C2 and EP1207392B1 (effective in Germany) cover the LMD PCR-Tube holder device with movable mounting elements for PCR-Tubes as collecting containers for dissectates

DE102005028062C5, EP1893971 and US8535876B2 protect the improved LMD cutting mode for fast cutting of dissectates.

EP1719998B1(effective in Germany, France and UK) and US7485878B2 protect the LMD fluorescence axis with fluo-cubes that are transparent for the laser beam and such allow active laser cutting in fluorescence modus.

US9217694B2 protects the ADM Software for LMD that provides the automated detection of dissectates and automated definition of cutting lines without the need of hand-input of marking lines
